Large-scale sequencing often aims at sequencing very long DNA pieces, such as whole chromosomes, although large-scale sequencing can also be used to generate very large numbers of short sequences, such as found in phage display. For longer targets such as chromosomes, common approaches consist of cutting (with restriction enzymes) or shearing (with mechanical forces) large DNA fragments into shorter DNA fragments. The fragmented DNA may then be cloned into a DNA vector and amplified in a bacterial host such as Escherichia coli. Short DNA fragments purified from individual bacterial colonies are individually sequenced and assembled electronically into one long, contiguous sequence. Studies have shown that adding a size selection step to collect DNA fragments of uniform size can improve sequencing efficiency and accuracy of the genome assembly. In these studies, automated sizing has proven to be more reproducible and precise than manual gel sizing. The term "de novo sequencing" specifically refers to methods used to determine the sequence of DNA with no previously known sequence. De novo translates from Latin as "from the beginning". Gaps in the assembled sequence may be filled by primer walking. The different strategies have different tradeoffs in speed and accuracy; shotgun methods are often used for sequencing large genomes, but its assembly is complex and difficult, particularly with sequence repeats often causing gaps in genome assembly.

Having told Neo that he now has the power to choose to end the war and defeat Smith, the Oracle then states "everything that has a beginning has an end, Neo" and informs him that in order for true peace to happen, he must travel to the heavily guarded Machine City in the real world and make a truce with the machines in order to save both races from extinction because of Smith's activities.
=== Musculoskeletal === Evidence does not support a benefit in delayed-onset muscle soreness. It may be useful for muscle pain and injuries. A 2008 Cochrane Library review concluded that LLLT has insufficient evidence for treatment of nonspecific low back pain, a finding echoed in a 2010 review of chronic low back pain. A 2015 review found benefit in nonspecific chronic low-back pain. LLLT may be useful in the treatment of both acute and chronic neck pain. In 2013, however, a systematic review and meta-analysis of LLLT for neck pain indicated that the benefit was not of significant importance and that the evidence had a high risk of bias. In a study testing the efficacy of low-level laser therapy treating plantar fasciitis found that LLLT significantly reduces pain in lower extremity tendinopathy and plantar fasciitis in the short and medium terms. The same study also stated that while comparing the effect of LLLT to that of therapeutic ultrasound in persons with patellar tendinopathy, and they found a statistically significant effect in favor of LLLT, both on pain reduction and function. There are tentative data that LLLT is useful in the short-term treatment of pain caused by rheumatoid arthritis, and possibly chronic joint disorders. Research that compared the effects of LLLT against other treatments, sham treatments, or no treatment at all, and randomized adult patients with rheumatoid arthritis to receive it were considered.
=== Monomers and repeat units === The identity of the repeat units (monomer residues, also known as "mers") comprising a polymer is its first and most important attribute. Polymer nomenclature is generally based upon the type of monomer residues comprising the polymer. A polymer which contains only a single type of repeat unit is known as a homopolymer, while a polymer containing two or more types of repeat units is known as a copolymer. A terpolymer is a copolymer which contains three types of repeat units. Polystyrene is composed only of styrene-based repeat units, and is classified as a homopolymer. Polyethylene terephthalate, even though produced from two different monomers (ethylene glycol and terephthalic acid), is usually regarded as a homopolymer because only one type of repeat unit is formed. Ethylene-vinyl acetate contains more than one variety of repeat unit and is a copolymer. Some biological polymers are composed of a variety of different but structurally related monomer residues; for example, polynucleotides such as DNA are composed of four types of nucleotide subunits.

=== Distillation and absorption packed beds === Distillation and absorption separation processes using packed beds for vapor and liquid contacting have an equivalent concept referred to as the plate height or the height equivalent to a theoretical plate (HETP). HETP arises from the same concept of equilibrium stages as does the theoretical plate and is numerically equal to the absorption bed length divided by the number of theoretical plates in the absorption bed (and in practice is measured in this way).
Teleosts have four major life stages: the egg, the larva, the juvenile and the adult. Species may begin life in a pelagic environment or a demersal environment (near the seabed). Most marine teleosts have pelagic eggs, which are light, transparent and buoyant with thin envelopes. Pelagic eggs rely on the ocean currents to disperse and receive no parental care. When they hatch, the larvae are planktonic and unable to swim. They have a yolk sac attached to them which provides nutrients. Most freshwater species produce demersal eggs which are thick, pigmented, relatively heavy and able to stick to substrates. Parental care is much more common among freshwater fish. Unlike their pelagic counterparts, demersal larvae are able to swim and feed as soon as they hatch. Larval teleosts often look very different from adults, particularly in marine species. Some larvae were even considered different species from the adults. Larvae have high mortality rates, most die from starvation or predation within their first week. As they grow, survival rates increase and there is greater physiological tolerance and sensitivity, ecological and behavioural competence. At the juvenile stage, a teleost looks more like its adult form. At this stage, its axial skeleton, internal organs, scales, pigmentation and fins are fully developed. The transition from larvae to juvenile can be short and fairly simple, lasting minutes or hours as in some damselfish, while in other species, like salmon, squirrelfish, gobies and flatfishes, the transition is more complex and takes several weeks to complete.

Azurin is a small, periplasmic, bacterial blue copper protein found in Pseudomonas, Bordetella, or Alcaligenes bacteria. Azurin moderates single-electron transfer between enzymes associated with the cytochrome chain by undergoing oxidation-reduction between Cu(I) and Cu(II). Each monomer of an azurin tetramer has a molecular weight of approximately 14kDa, contains a single copper atom, is intensively blue, and has a fluorescence emission band centered at 308 nm. ==== 2010s ==== In 2012, Starbucks had annual Frappuccino sales of over US$2 billion. In August 2012, the largest Starbucks in the US opened at the University of Alabama's Ferguson Centre. On June 25, 2013, Starbucks began to post calorie counts on menus for drinks and pastries in all of its U.S. stores. In July 2013, more than 10% of in-store purchases were made on customers' mobile devices via the Starbucks app. The company once again utilized the mobile platform when it launched the "Tweet-a-Coffee" promotion in October 2013. Filtering helps stabilise the flavour of beer, holding it at a point acceptable to the brewer, and preventing further development from the yeast, which under poor conditions can release negative components and flavours. Filtering also removes haze, producing a clear beer with a "polished shine and brilliance". Clear beer became commercially desirable with the increasing use of glass drinking vessels and the development of pale lagers. During lagering, suspended material settles out, allowing the beer to "drop bright" and develop a naturally clear appearance. There are several forms of filters; they may be in the form of sheets or "candles", or they may be a fine powder such as diatomaceous earth (also called kieselguhr), which is added to the beer to form a filtration bed which allows liquid to pass, but holds onto suspended particles such as yeast. Filters range from rough filters that remove much of the yeast and any solids (e.g., hops, grain particles) left in the beer, to filters tight enough to strain colour and body from the beer. Filtration ratings are divided into rough, fine, and sterile. Rough filtration leaves some cloudiness and some character in the beer, while Fine filtration removes almost all cloudiness and microorganisms. Polysaccharides, proteins, yeast, bacteria and the brewing process may affect filtration.

== Outlook == The long-term course of MCTD may vary. Long-term follow-up studies have shown that MCTD can progress to a moderate disease with a favorable prognosis, or patients can acquire a significant condition with vascular alterations driven by pulmonary hypertension and increased mortality. Approximately one-third of people with MCTD have a benign course and go into remission, while the other one-third have a more aggressive course with a poor response to treatment. Approximately one-third of MCTD patients improved with immunosuppressive medication but continued to require immunosuppressive therapy after several years. The prevalence of pulmonary hypertension was related to the worst prognosis and a high mortality rate, making it the most significant complication in MCTD.
